Mech:
An antimetabolite
Inhibits nucleotide synthesis (thymidine)

A folic acid analog

Tox:
Myelosuppression (*reversible with leucovorin)

Solid tumors, colon cancer, basal cell carcinoma (topical)
Synergy with MTX.

Inhibits thymidylate synthase
A pyrimidine analog (5F-dUMP), which complexes with folic acid

Tox:
Myelosuppression (*not reversible with leucovorin)
Treat and OD with thymidine
Photosensitivity.
