Hb converted to hemiglobin cyanide (HiCN) - [ ] measured by spectrophotometry

 

1.Blood dissolved in K ferricyanide and K cyanide (Hb oxidized to Hemiglobin (Hi;methemoglobin) and then converts to Hemiglobin cyanide (HiCN)

 

2. Absorbance at 540 nm reflects amount of hemoglobin originally present.
