Phosphorylation of drug by viral kinases leads to inhibition of DNA synthesis in virus-infected cells. Acyclovir, valacyclovir, famciclovir, penciclovir, ganciclovir, and valganciclovir are phosphorylated by viral kinases and then inhibit viral DNA polymerase. Cidofovir is phosphorylated by cellular enzymes and then inhibits CMV DNA polymerase.

The anti-HIV nucleoside analogues are phosphorylated by cellular kinases, and then inhibit viral reverse transcriptase. Anti-HBV nucleoside analogues are also phosphorylated by cellular enzymes, and then inhibit HBV polymerase.
