Mechanism, pkin/ddi

Definition

Mech: Block recycling of PIP2, especially works in brain. Also inhibits release of norepi, dopamine, increases 5-HT synthesis, uncouples G-proteins, alters electrical gradients

Pkin - ion, so excreted renally.Diuretics, ACEi inhibit excretion so increase concentration, slow onset of action

Definition

Nausea, diarrhea, acne, drowsiness, polyuria, polydipsia, weight gain.

Tremor, ataxia, aphasia, bizarre movements, confusion at high conc - renal impairment

Intereferes w/vasopressin - nephro diabetes insipid

Interferes with TSH

Some badness w/pregnancy
