Cards

Term
CITRATE AGAR
Definition
- tests for citrase activity

- citrate --> oxaloacetate via citrase

- color change from green --> blue (acidic --> basic)

- blue = Positive for citrase activity
Term
Sulfide Indole Motility (SIM) Agar
Definition
- tests for H2S, motility, indole

- H2S production characterized by formation of black ppt.

- motility: if turbid, yes. if not, no

- indole production: add Kovac's reagent; if red ring forms, then (+) for indole (tryptophan --> indole by bacterial enzyme tryptophanase)
Term
Motility Indole Ornithene (MIO) Agar
Definition
- tests for indole, motility, and ornithene decarboxylation

- motility: measured by turbidity

-ornithene decarboxylation: if purple, then (+) (BASIC); if yellow, (-) acidic

- Indicator = brom-cresol purple

- indole = red ring means (+) for indole
Term
Urea Broth
Definition
- tests for urease activity

- (NH2)2CO (urea) --> CO2 + H2O + NH3 (ammonia)

- color change from yellow to bright pink indicates that urease is present

- yellow (acidic) --> pink (basic)
Term
Carbohydrate Broths (glucose, sucrose/dextrose/lactose)
Definition
- tests for carbohydrate fermentation into acidic end products: CHO --> OH- (acidic) + CO2

- indicator = phenol red

- yellow = (+) for acidic end products, red = (-)
Term

Methyl Red Broth

Definition
- tests for ACIDIC END PRODUCTS of glucose fermentation

- glucose (ferm)--> OH- (acidic) + CO2

- red = (+) for acidic end products
Term
Voges - Proskauer Broth
Definition
- tests for NONACIDIC END PRODUCTS of glucose fermentation

- glucose (utilization) --> non-acidic end products

- pink = (+) for non-acidic end products, yellow = (-)
Term
Nitrate Broth
Definition
- tests for nitrate reductase activity

- NO3(2-) (nitrate) --> NO2(-) --> N2(g)

- test by adding reagents A & B --> if color change, (+) for nitrate reductase; if no color change, add reagent C(zinc)--> if color change then, (-) for nitrate reductase
