Definition
Rate of protein movement. Dye a cell fluorescent, bleach part w/laser to kill the proteins there, see how quickly it fills back up w/fluorescent proteins.

Fluorescence resonance energy transfer (FRET)
Definition
To see if proteins interact. Dye 1 protein w/specific fluorescence, dye second protein w/other specific fluorescence. First protein's emission excites the second protein so if the second protein glows they associate.

2D gel electrophoresis
Definition
Use a pH separation in addition to a size separation to separate in the Y axis by size and the X axis by charge. Resolves a few thousand proteins, no longer in use.

Definition
Digest protein sample w/protease. Ionize peptides in a mass spectrometer, obtain mass spectrum, use computer to match spectrum w/database values.

Shotgun mass spectrometry
Definition
Digest protein MIXTURE w/trypsin, fractionate peptides w/liquid chromatography and electrospray ionization. Randomly select peptides from first MS, collision-induced fragment (CID) the peptides, and analyze again to get a smaller complex's mass spec. Use computers for database search.

Label 2 cultures w/either heavy or light amino acids. Mix proteins. Peptides will form layers based on weight, amount of layer = amount of that protein.
