HMG-CoA reductase inhibitors:
Most effect comes from Hepatic uptake of LDL in response to inability to synthesize it.

Number one antihyperlipidemic (LDL mainly followed by VLDL)

Rosuvastatin>Atorvastatin>Simvastatin

Also works as an antioxidant/antinflammatory

DO NOT GIVE PREGNANT WOMEN

May cause rhabdomylysis

Modest Hepatoxicity

Moderate increase in Type II diabetes

Big risk factor is CYP450 inhibitors such as anti-HIV drugs

OAT1b1 polymorphism increases blood lvls and rhabdomylysis/K increase

Cause myopathy with statins
