Term
what is a SIDE EFFECT?
Definition
Unwanted, non-deleterious effects appearing after administration of standard therapeutic doses and RELATED TO THE PHARMACOLOGICAL PROPERTIES of the drug
Term
what are some FEATURES of drug SIDE EFFECTS?
Definition
-found with most drugs, but patients are variable, generally not serious
-dose dependent and related to drug properties
-mechanism often drug receptor interaction
-switching drugs or changing dose is recommended
Term
what is OVERDOSE TOXICITY?
Definition
Unwanted effects appearing after administration of doses higher than the therapeutic ones
Term
what are the FEATURES of OVERDOSE TOXICITY?
Definition
-all drugs, all subjects (if dose is high enough)
-danger can be high depending on drug and dose
-drug receptor interaction
-treated with antagonist or lower dosage
Term
what is a DRUG IDIOSYNCRACY?
Definition
A genetically determined, abnormal reactivity to a drug
Term
FEATURES of DRUG IDIOSYNCRACY?
Definition
-low incidence, depending on person and drug
-can be serious depending on drug and dose
-stop drug and use antagonist if available
Term
what is a DRUG ALLERGY?
Definition
An unwanted drug effect resulting from previous sensitization to the same drug or a closely related one
Term
FEATURES of a DRUG ALLERGY?
Definition
-low incidence despite possibility in many drugs
-can be very serious
-unlike others which are drug receptor dependent, this is IMMUNOLOGICAL in mechanism
Term
what are the 4 types of DRUG ALLERGIES?
Definition
TYPE I - immediate rxns--> via IgE of mast cells and basophils
TYPE II- antibody dependent cytotoxicity-->drug binds to self cell eliciting immunological response
TYPE III-immune complex-mediated reaction
Type IV-delayed or cell-mediated reaction (contact dermatitis is most common)
