Tox: OD produces NAPQI → hepatotox

Antidote: N-acetylcysteine

Bisphosphonate (others: pamidronate, alendronate, risedronate)

Mech: inhib osteoclasts → decr. formation & resorp of hydroxyapatite

Tox: corrosive esophagitis

Anti-gout

Mech: binds & stabilizes tubulin → inhibs MT polym → impairs WBC chemotaxis & degranulation

Use: Acute gout

Tox: GI (indomethacin used more often d/t SE)

Anti-gout

Mech: blocks OAT → inhibs reabs of uric acid in PCT

Use: chronic gout (not acute)

Interact: inhibs secr. of Pen in PCT

Can cause uric acid stones (hyperuricurea)

Anti-gout

Mech: inhibs xanthine oxidase → decr. xanthine=>uric acid

Use: chronic gout (not acute); prevention of tumor lysis urate nephropathy in lymphoma/leukemia

Interact: incr. conc of azathioprine & 6-MP (degraded by XO)

Mech: recombinant TNF R that binds TNF

Use: RA, psoriasis, AS

Mech: anti-TNF Ab

Use: Crohn's, RA, AS

Tox: predisposes to infxn (reactiv. of TB/ histo)
