| Make POB and Glucose control plates and put ADP 6 and 1 into both. Put 1.5 mL ADP-1 in centrifuge for 2 min at 10k RPM. Discard supernatant, keep pellet. Put 300 microL TENS to pellet to lyse cells. Make serial dilutions. Prepare a POB plate with ADP-6 with spread plate technique. Divide to 4 quadrants. In quadrant 1, put 1 microL ADP-1 serially diluted by 10 --> quadrant 4, serially diluted by 10^(-3). Grow for 24 to 48 hrs. |
