Term
| MRVP Broth Reading Results |
Definition
Tube 1: add ½ dropperful of alpha naphthol, and 40% potassium hydroxide Cap tightly & shake vigorously, wait 10 minutes, shake again. Red = +voges proskauer (AMC), all other colors are negative
Tube 2: add ½ dropperful of methyl red… red result is MR+, orange is –test |
Term
| Eosin Methylene Blue (EMB) |
Definition
-suppresses the growth of g+ while allowing the growth of g- - G - that grow, this agar differentiates between those that ferment lactose and those that do not.Lactose fermenters=darker than the agar itself. (E.coli shows up metallic green.) |
Term
Definition
| differential and enriched medium that is a TSA base with 5% sheep’s blood to enhance the growth of pathogens. Differential medium based on the degree of hemolysis of RBCs in the agar. Be sure to make evaluations where colonies are sparse and isolated. Heavy thick growth displays ambiguous readings. |
Term
| Blood Agar Reading Results |
Definition
gamma hemolytic – no lysis of blood in agar alpha hemolytic – partial or incomplete lysis of red blood cells in agar appears as a greenish, distinctly bordered opaque zone around each colony beta hemolytic – complete RBC lysis appears as clear, distinct opaque borders around each colony |
Term
Definition
| selective and differential medium used to isolate bacteria from stool cultures. This medium isolates most common intestinal pathogens. G+ bacteria are inhibited while Salmonella and Shigella species show up lightly colored or black-centered on this agar. Common non-pathogen coliforms (E. coli) show up brightly colored. |
Term
Definition
| selective and differential medium designed to isolate skin organisms and high salt tolerant organisms such as Staphylococcus. The selective factor is high salt concentration. The differential feature is the fermentation of mannitol sugar. The phenyl-red acid/base indicator turns yellow in low pH indicating fermenters while red or hot pink indicates neutral to high pH of non-fermenters. |
Term
Definition
| differential and enriched medium contains cooked blood and tellurium based salts. The salts differentiates those bacteria that can metabolize the tellurite. The blood enhances the growth of organisms such as Corynebacterium (for which the agar is designed). Black colonies occur with tellurite reducers. |
Term
Definition
| differential medium that selects for various species of Pseudomonas species. The agar accelerates the production of fluorescent pigments. This fluorescence is examined under UV light after incubation. |
Term
Definition
| agar displays fermentation of three sugars (triple) that this medium contains…glucose, sucrose, and lactose. Fermenters turn the phenyl-red acid/base indicator to yellow. Non-fermenters cause the medium to remain red. This medium also indicates amino acid metabolism by the production of hydrogen sulfide as a by-product. This is indicated by the formation of a brown-black precipitant when the hydrogen sulfide reacts with the iron in the medium. |
Term
Definition
| culture medium that allows the growth of certain types of organisms, while inhibiting the growth of other organisms |
Term
Definition
| culture medium that allows one to distinguish between or among different microorganisms based on a difference in colony appearance (color, shape, or growth pattern) on the medium. |
